do not vortex

You state here not to vortex the sample after addition of tri* and chloroform. What evidence do you have for that? The TRIZOL manual, for example, does not take particular care here: "mix the sample by vortexing".

  • Jakob, interesting question, the answer to which has faded with the passing of time. The current Tri Reagent protocol from MRC states "shake vigorously for 15 seconds", and doesn't exclude or specify vortexing. To be honest I don't know where the "do not vortex" on our protocol came from - it may be a carry-over from an ancient Tri protocol, or urban myth, Ajeffs 14:27, 12 February 2008 (CST).
