Questions for Stephen

  1. Is the sequence of the J45017 reverse primer correct? It seems quite short.
    • A: I think I checked both the tube and the wiki, but I can doublecheck tomorrow.
      • Double-checked... is correct... I remember it was very short because of its high GC content
  2. Did you use the Knight or Endy site directed mutagenesis protocol? (http://openwetware.org/wiki/IGEM:MIT/2006/Notebook/2006-8-10#Mutagenesis says Knight but you told me Endy)
    • A: We used the Knight protocol.
  3. Quantification of banana timecourse
    • A: I sent both Alex and Amy an email today about a meeting tomorrow.
      • You might want to do some background research on quantification of GC data.
  4. The reverse primer for <bbpart>BBa_J45992</bbpart> listed in the google doc does not match what is listed in the Registry
    • A: I will double check this, but I'm pretty sure that I confirmed this via the wiki and Isadora's order sheets. I do not know who wrote the Registry page (maybe Andre), but I do trust the lab notebook more than the Registry.
      • OK then the Registry page needs to be fixed after you confirm.

  5. The forward and reverse primers for <bbpart>BBa_J45993</bbpart> listed in the google doc does not match what is listed in the Registry

  7. <bbpart>BBa_J45992</bbpart> and <bbpart>BBa_J45994</bbpart> need to be submitted to the Registry. Or at least the GFP versions do.
  8. Does <bbpart>BBa_J45017</bbpart> still have the PstI site or has it been mutated out? If it has been mutated out, then the Registry needs to be updated.
