Questions for Stephen

  1. Do we have the regulated wintergreen GC data in a form that we could generate plots similar to the banana timecourse?
    • Yes, we could. It would take some digging through old files and sorting out through each protocol we used on each day. I don't know if it's necessary to show though.
      • Drew would like to show these data if we can.
  2. What did you use to shake the cultures? i.e. what incubator or shaker?
    • 220 RPM- the shaker in 37 C room
    • 110 RPM- the shaker by Samantha's/2007 iGEM team's bench
  3. What concentration salicylic acid and isoamyl alcohol did we use for the scent generator tests? Was it always 440 ppm isoamyl alcohol?
    • A: Always 440 PPM isoamyl alcohol, 276 PPM salicylic acid
      • Are you sure of your math here? You told me in an email that the cultures for the banana data had 0.4mM isoamyl alcohol but in the spreadsheets, you say 4mM. But 440 PPM of isoamyl alcohol is 5 mM isoamyl alcohol by my calculations. 276 ppm salicylic acid is 2mM salicylic acid
  4. A summary of your analysis of fixing the wintergreen precursor production would be useful
  5. Are J45900 and J45700 on a single plasmid each? Just confirming
