(April 28; MDolinar) Hi, Joseph. You mentioned the major band on the gel is chromosomal DNA. This doesn't sound like a very useful method for plasmid DNA isolation then... We use the one-tube miniprep method according to Del Sal et al. (NAR 16, 9878; 1988). Essentially, it is much like the published OWW protocol, just that it includes a CTAB DNA precipitation step prior to ethanol ppt. Regards, Marko
