Cancer is one of the most malignant diseases, which accounts for about 13 percent of deaths in the world. One of the most effective approaches for it is chemotherapy, but it often attacks not only cancer cells but also normal cells, leading severe side effects. Recently the drug delivery system has been developed to improve cancer-specificity, but it isn't easy to design delivery systems one by one according to each drug. </p>
 <p class="paragraph">
Here we propose an anti-cancer drug which doesn’t need delivery system for cancer specificity: cancer-specific pore-forming DNA origami drug. Inspired by a toxin produced by an animal and a mechanism existing in human immune system, we designed DNA origami subunits which coordinately kill a cell. The process of killing is as follows. Subunits stick in the cell membrane with cholesterol site. Then they oligomerize so that they form pore on the membrane. This pore disrupts the phospholipid bilayer of cells, leading cell lysis and death. Since we use DNA origami, it is expected to be possible to combine DNA origami computing circuit of the past research with it and improve cancer-specificity.

In Japan meeting for biomod2013, we will report the interaction between DNA nanostructures and cell membrane and several methods for oligomerizing.   </p>
