Objective

  • To create standard solution of gold for calibration of the atomic absorption spectrometer. The students will measure the gold content of their BSA nucleated AuNPs against these standards.

Bench Work

  • 1000 ± 10μg of Au/mL of H2O stock solution
    • from Inorganic Ventures cat# AAAU1-1, lot # F2-AU04012
    • the solution is certified by ICP (ion couple plasma spectroscopy) to NIST SRM 3121

For the AAS (atomic absorption Spectrometer), the published detection limit for gold is 0.2 ppm ( a 1% absorbance)for this instrument. recent experiment is closer to 2 ppm and since 20 ppm is roughly 0.1 mM Au, Therefore I created standards ranging from 5 to 40 ppm.

  • created 50 mL of each standard: 5, 8, 10, 15, 20, 25, 30, 40 ppm Au from the 1000 ppm Au stock standard.
ppm mL of 1000 ppm stock added
5 0.25
8 0.40
10 0.50
15 0.75
20 1.00
25 1.25
30 1.50
40 2.00
