* Synthesize lysozyme-AuNPs with salt added before being heated.
* Image tests from yesterday.
* Prepare diluted solutions of citric acid, MES, 2,2 bipyridine, NaCl, KCl, MgCl<sub>2</sub>, CaCl<sub>2</sub> from 1 M to 100 mM, 10 mM, and 1 mM.
* Experiment with various concentrations to determine the minimum concentration of citric acid, MES, 2,2 bipyridine, NaCl, KCl, MgCl<sub>2</sub>, CaCl<sub>2</sub>, HEPES, and Tris that causes fiber formation.

Objective

  • Synthesize lysozyme-AuNPs with salt added before being heated.
  • Image tests from yesterday.
  • Prepare diluted solutions of citric acid, MES, 2,2 bipyridine, NaCl, KCl, MgCl2, CaCl2 from 1 M to 100 mM, 10 mM, and 1 mM.

Procedure

Creating Stock Solutions of Gold and Lysozyme

  • As seen below, each stock solution was created following the calculations in the table.
  • Each solution was created using a volumetric flask and deionized water.


Synthesizing Lysozyme-AuNPs

  • As seen below, each solutions was created using the following calculations.
  • We made one tube of each ratio for the following 1 M salt solutions: KCl, NaCl, CaCl2, MgCl2


Adding Diluted Variables to the Lysozyme-AuNP Samples

  • Dilute the stock solutions of citric acid, MES, 2,2 bipyridine, NaCl, KCl, MgCl2, and CaCl2 to 100 mM, 10 mM, and 1 mM.
  • Add 0.5 mL of each variable to the 30, 55, and 110 ratios of lysozyme-AuNPs.
