Tasks for January 27

  • Formation of 30:1 Gold-Lysozyme nanoparticle colloid
  • Formation of 45:1 Gold-Lysozyme fibers

Preparation of stock solutions

  • 100 mL of 2.512mM HAuCl4 stock solution was prepared by adding 0.085 g of HAuCl4 in distilled water.
    • (0.1L)(2.512×10-3 mol/L)(340 g/mol)=0.085 g (0.08533 g were weighed out)
  • 50 mL of 20 μM lysozyme stock solution was prepared by adding 0.014307 g of Lysozyme in distilled water
    • ((0.05L)(20×10-6 mol/L)(14307 g/mol)= 0.014307 g (0.01417g were weighed out)

Note: The pH of the solution has to be kept at 3.6 for optimum formation of AuNPs.

Preparation of Au colloid and fibers

30:1 AuLys colloid

  • Per mL test tube, add 0.5 mL HAuCl4 stock solution (final concentration 251 μM)
  • Also add 2.095 mL of 19.81μM of lysozyme stock
    • m1v1=m2v2
    • 8.3×10-6(5mL)=19.81×10-6v2
    • v2=2.095 mL
  • Make up to 5 mL with distilled water (2.405 mL)
  • Repeat for 9 other test tubes

45:1 AuLys fibers

  • Per mL test tube, add 0.5 mL HAuCl4 stock solution (final concentration 251 μM)
  • Also add 1.40 mL of 19.81μM of lysozyme stock
    • m1v1=m2v2
    • 5.56×10-6(5mL)=19.81×10-6v2
    • v2=1.40 mL
  • Make up to 5 mL with distilled water (3.1 mL)
  • Repeat for 9 other test tubes

Place in oven for 4h at 80°C.
