We may have tweezing... yay!

Tethering

Setup

  1. Clean glass: 180sec in sonicator, rinsed thoroughly, blow dried
  2. bead prep: 1:25 dilution in BGB
  3. DNA: pBR322 diluted 1:10 from gel extracted amount

Results

Tethering was pretty smooth. There were tons of tethers although at first, there wasn't that many. The solution of beads was a little more clumpy than I had hoped for, but I guess I should sonicate a little more next time. I tried unzipping, but I don't really understand the data yet so who knows what I got.
