Notes

Things I've done:

  • made new beads diluted 1:100 in 1mg/ml dephosphorylated a-casein
  • 1mg/ml dephos a-casein in 1x Popping buffer
  • made 4.4kb stretching DNA
    • 1:100 dilution in popping buffer

Tethering things I've tried recently with 1um beads:

  1. BGB washes with BGB beads
    • nearly no tethers, possibly from bead-glass surface interactions
  2. concentrated anti-dig with d-a-casein washes with d-a-casein beads
    • beads stick to glass
  3. anti-dig with d-a-casein washes with d-a-casein beads
    • beads stick to glass
  4. anti-dig with d-a-casein wash (1 time) and subsequent washes in pop with d-a-casein beads
  5. anti-dig with BGB wash (1x) and subsequent washes with pop with d-a-casein beads
