Things I've done:
- made new beads diluted 1:100 in 1mg/ml dephosphorylated a-casein
- 1mg/ml dephos a-casein in 1x Popping buffer
- made 4.4kb stretching DNA
- 1:100 dilution in popping buffer
Tethering things I've tried recently with 1um beads:
- BGB washes with BGB beads
- nearly no tethers, possibly from bead-glass surface interactions
- concentrated anti-dig with d-a-casein washes with d-a-casein beads
- beads stick to glass
- anti-dig with d-a-casein washes with d-a-casein beads
- beads stick to glass
- anti-dig with d-a-casein wash (1 time) and subsequent washes in pop with d-a-casein beads
- anti-dig with BGB wash (1x) and subsequent washes with pop with d-a-casein beads
