  • Cloning:
    • We ran a gel on our pcr products from yesterday (Fra 1 antisense and sense extracted from fresh strawberries)
      • Based on the gel, it doesn't seem that the sequences were amplified (Wanted ~ .3 kb and saw .1 kb)
      • Reasons: primer dimers, cDNA extraction, allergen not expressed in particular strawberry
    • Digest w/ Xbai1 and Pst1/ purify to ligate into V0120 (biobrick plasmid)
    • Transform into E.Coli
  • Artificial miRNAs
    • It seems that we will probably create artificial miRNAs for silencing
    • We are waiting on the intron from the pHannibal vector
