Objectives

  1. Work on Disc Diffusion Antibacterial Procedure and Prepare Reagents
  2. UV Vis on 02/24/16 Nanoparticle Reactions

Materials

  1. Ocean Optics UV Vis was used to record UVVIS Spectra of Casein AgNPs. The other machine broke and is unusable.
    1. Integration time= 10ms and Average =100.
    2. A water blank in the same quartz cuvette was subtracted from the spectra before running any samples.

Procedure

  1. Materials for Agar Disc Diffusion
    1. Mueller Hinton Agar
      1. Dissolved Agar packet in 1L of water over hot plate
      2. Autoclaved on wet cycle
  1. Reaction mix data can be found at the link SRB Lab Notebook Data

and the reaction itself was run on 02/24/16, the reaction mix planning and UV Vis from 02/17/16 was performed on the 02/23/16

Results

  1. Color change notes and UV-VIs information can be found at the link SRB Lab Notebook Data
  1. It was noted that diluting Pea Protein in base allowed for the silver nanoparticle reaction. From now on, will dilute pea protein in base. This is similar to what was experienced with brown rice protein and casein - both of which were originally dissolved in base and synthesized silver nanoparticles.
