New PCR

New PCR today. It is essentially the same as the last couple days, but the reverse primer is changed to R2008.

Since both Anthony and I are both satisfied with with last weeks results, we are moving on. This reaction is testing the exact same thing as before, the affect of the concentration of Mg2+ cations on the PCR. Going across the table, the concentration increases from 1.0 mM to 5.0 mM in 0.5 mM increments, as indicated by the number of the tube. We are also running the annealing step at 60°C as apposed to the 50°C that we did for the second round of PRC from last week. The gel and results from the reaction will be posted tomorrow.
