Objective

Analysis and synthesis of more protein-AuNP materials. Each group will have a different task. One (set) of groups will be doing AA on their samples from last weed (HRP-AuNPs and pepsin-AuNPs) and this week (Myoglobin-AuNPs and HRP-AuNPs). Another (set) will take UV-Vis absorbance of their samples (this week and last week). The final (set) will synthesize new protein-AuNPs.

Procedure

Creating the Gold Stock Solution

  1. Add 0.0506 g of HAuCl4·3H2O were added to a 50 mL volumetric flask and filled with distilled water.
  2. The final concentration of gold stock was 2.57 mM.

Creating the Lysozyme Solution

  1. As seen in Figure 1, the corresponding amount of protein was added to a 50 mL volumetric flask and filled with distilled water. The samples were made in small test tubes and capped with aluminum foil.
  2. The samples were heated at 80°C for 4 hours.

Notes

  • The final gold concentration in the solutions was 0.25 mM.
