Objective

Synthesize Au nanoparticles

Description

  1. Create a 10 mL aqueous mixture of BSA (0.0015 - 0.015 mM) and HAuCl4 (0.25 - 1.0 mM)
  2. Place the mixture in a water thermostat bath at 80°C
  3. Measure the reaction progress every 30 minutes using a UV-vis Spectrometer with a wavelength range between 200 and 800 nm. The reaction should be at equilibrium when the peak at 550 nm reaches a maximum intensity.

  • A graph of the corrected absorbance over time was constructed by taking a spectrum of the buffer was taken and that absorbance was subtracted from the absorbance taken of the sample every half and hour:

  • a graphof the corrected absorbance over wavelength zoomed in on the 550 nm wavelength.

  • This graph show the absorbance increasing over time which indicates that gold nanoparticles were being formed in the reaction mixture as the gold was being reduced:

Notes

  • 0.001 g of BSA used, 0.001 g HAuCl4 measured and a little less used in tube
  • Took a UV spectra at t=0. Oven was heated to 78C when sample was placed inside. Placed inside at 12:04 pm.
  • Took a UV spectra of blank with buffer solution 50 mM Acetate Buffer pH 3.6
  • Took UV spectra at 1=30. Oven heat at 69C when sample was placed inside. Placed inside at 12:41 pm
  • Took UV spectra at t=60. Oven heat at 78C when sample was placed inside. Placed inside at 1:16 pm
  • Took UV spectra at t=90. No change. Oven heat 70C when samples was placed inside. Placed inside at 1:50 pm.
  • Took UV spectra at t=127. No change. Oven heat 70 degrees when sample was placed inside. PLaced inside at 2:38.
  • Took UV spectra at t=157. No change. Oven heat 70 degrees when sample was placed inside. Placed inside at 3:12.
  • Took UV spectra at t=187. Wrapped in foil and set at room temperature.

Calculations: Au: 0.25mM Au = 0.00025 mol / L = 2.5 x 10-7 mol / mL 2.5 x 10-7 mol / mL = x / 10 mL x = 2.5 x 10-6 mol / mL x 196.967 g/mol = 4.92 x 10-4g BSA: 0.0015mM BSA = 0.0000015 mol / L = 1.5 x 10-9 mol / mL 1.5 x 10-9 mol / mL = x / 10 mL x = 1.5 x 10-9 mol / mL x 66776 g/mol = 1.00 x 10-3g

  • After a week the solution turned a light purple
