Entry title

Objective

  • Extracting Protein from cell cultures

Description

Extraction

  1. Defrost cells that were stored at -20°C.
  2. The cells were sonicated for 15 seconds at a time with approximately one minute on ice between sonications. In total the cells were sonicated for 90 seconds.
  3. The cells were centrifuged at 9000g for 30 minutes at 4°C.
  4. Purify supernatant using syringe filter
  5. Store purified supernatant in refrigerator

Perform Bradford Assay

  1. test each fraction using Bio-Rad Protein Assay using Bradford "by-eye"
