HEK293 Luc #4 fixed DAPI staining

  • DAPI/Hoescht: made by Dr. Haynes; 5000x in sterile dH2O
  • Diluted Hoescht to working concentration (1x) in 1xPBS.
  • Fixing cells with formaldehyde
  • Adding Hoescht to the cells, let sit in for 10 mins in the dark to develop signal

Results:

  • Good DAPI signal
  • Detection Algorithm failed due to very high cell density

Next steps:

  • Growing and imaging cells in less dense conditions
