The Daily Grind

  • To do today:
    • Plate out all soil plant endophyte glycerol stocks and incubate for 10 days (dilute 10 ul glycerol in 90 ul water and spread)
    • Gel purify digested PCR and plasmids and ligate overnight
    • Extract DNA from all the frozen soil plant samples and spec
    • Analyze second rep of TRFLPs from seeds

The E. coli survival experiment will have these specs:

Two E. coli - Empty BAC 2O2 and Initially positive BAC 1O1 Three dip treatments - colony resuspention in LB, O/N in LB, and O/N in LB with antibiotics Sampling of the entire plant by grinding and plating on LB + chloramphenicol + XGal. Two reps of this. Sampling times will be Just after dipping, Day 1, Day 3, Day 7, Day 14. OD600 of all innoculants will be taken
