Remains of the Day

  • Continue 2 hour EtOH vs overnight EtOH trial - used 15 ul of mixed PCR in each of these, and 0.25 ul of DdeI per tube with a 16 hr overnight incubation at 35 degrees.
  • Start strawberry soil trial extraction (how many reps should we extract to get robust TRFLP results?)
  • Restriction enzyme selection for ITS1F/ITS4 PCR : http://aem.asm.org/content/75/14/4747.full - buying BstNI for this, cause its amongst the best and is the cheapest for what it is.
