Remains of the Day

  • Spray tomatoes
  • Do digests of PCRs from yesterday, loading same total amount of DNA (calculate amount of enzyme needed and do two digests - overnight and intraday)
  • Review grant and insert new references
  • Did gradient PCR at 48, 51, 56 and 60 degrees using 8 of Jons good soil, bad soil samples. Used 2 ul of each "diluted" sample per 50 ul PCR. Ran a set with 799f/1492rh (10 pmol instead of the 20 used around here) at 50 degrees
