Goal: To develop endophytic microbial biofertilizers for corn agriculture.
Objective 1: Survey natural varation of microbes found inside corn seed and plants.
A) Survey corn seed endophytes.
B) Survey soil transmitted endophytes from Mexico.
C) Search for growth promoting endophytes through seed innoculation bioassay.
D) Gain an understanding of endophytic mechanisms of plant growth promotion including colorimetric biochemical tests and GFP tagging of bacteria to ensure endophytic localization in planta.
Objective 2: Genetically modify endophytic microbes to improve their growth promoting properties.
A) Modify Klebsiella pneumonii to transgenically produce ACC deaminase and acetoin and assay its effect on plant growth.
B) Isolate genes involved in plant growth promotion in the microbe, G. azotocaptans.
