Reaction of OPD+H2O2 with Mb Catalyst in Acetone Suspension

The procedurefor the reaction of OPD and H2O2 was followed using a Mb/Tris catalyst and an acetone suspension. The reaction was run in a beaker with a stir bar and the solution was centrifuged for 1 minute at 13,200 rpm before taking a spectrum.

In addition reactions were run using Mb/Phosphate and Mb/Acetate in hexanes suspension. These samples were filtered (not centrifuged) before taking a spectrum.

Mb/Tris in Acetone, Trial 1

The drop in absorbance at 140 minutes is due to the outside of the cuvette being cleaned.

Mb/Tris in Acetone, Trial 2

The drop in absorbance at 120 minutes and 140 minutes is due to the outside of the cuvette being cleaned.

Mb/Phosphate in Hexane

Mb/Acetate in Hexane

Note: The high absorbance peak at 140 minutes is due to a potential water/protein precipitate that filtered through.
