High Stability Plasmid Development

Abstract

  • Replating of colonies obtained for the transformations of August 30

Replating

Transformations were successful, except for P1001 (Tetracycline resistance in plasmid pSB1AK3). I replated several bacterial colonies in new petri dishes:

B1007 Bidirectional Terminator Am, Kn 8
B1001 Bidirectional Terminator Am, Kn 2
B0014 Bidirectional Terminator Am, Kn 8
P1004 Chloramphenicol resistance Am and Am, Cm 5


Two colonies from each transformation were chosen to be inoculated in 5mL of liquid LB with 5μL of the proper antibiotic for plasmid extraction tomorrow.
