Objective

To try to stop the unfolding process of BSA and to determine in what situations BSA/AuNP fibers are stable.

Description

  1. Make two solutions of AuNP.
    • Solution 1 (aqueous AuNP):
      1. Combine 0.516 mL of BSA and 0.350 mL of HAuCl4 in a test tube.
      2. Place in 80°C oven until solution turns purple.
    • Solution 2 (AuNP/BSA fibers):
      1. Combine 0.516 mL of BSA and 0.700 mL of HAuCl4 in a test tube.
      2. Place in 80°C oven for 2 hr.
  2. Remove solution 1 from oven.
  3. Add 100 μL of 5.02 mM Tris (pH 10.1) to 1 mL of solution 1.
  4. Take UV vis data.
  5. Repeat steps 3 and 4.

Notes

Solution 1 began to turn purple after 1.5 hr in the oven, at which point it was removed.

Small purple fibers formed in solution 2.
