Objective

To try to stop the unfolding process of BSA and to determine in what situations BSA/AuNP fibers are stable.


Also, to repeat the experiment done on 11/15/11.

Description

  1. Make two solutions of AuNP.
    • Solution 1 (aqueous AuNP):
      1. Combine 0.516 mL of 30 mM BSA and 0.350 mL of HAuCl4 in a test tube.
      2. Place in 80°C oven until solution turns purple.
    • Solution 2 (AuNP/BSA fibers):
      1. Combine 0.516 mL of 30 mM BSA and 0.700 mL of HAuCl4 in a test tube.
      2. Place in 80°C oven for 3.5 hr.
  2. Remove 1 mL of solution 1, place remainder back in oven.
  3. Take UV vis data.
  4. Add 100 μL 50.1 mM tris buffer (pH 10.1).
  5. Repeat steps 3 and 4.
  6. After 1.5 hr, remove solution 1 from oven again.
  7. Take UV vis data.
  8. Add 100 μL 50.1 mM tris buffer (pH 10.1).
  9. Repeat steps 7 and 8.
