Objective

Today's objective is to synthesize Gold NPs; we want to obtain a purple solution and fibers using Gold/BSA ratio we determined last semester. These solutions will then be used next week to test the binding of the dye.

Protocol

  • Gold/BSA ratio = 70
  1. Solution A: 0.042mL of HAuCl4 (2.9mM) + 0.1mL of BSA (17.7µM) + 0.85mL of water (2hours in the heatblock at 80°C)
  2. Solution C: 0.42mL of HAuCl4 (2.9mM) + 1mL of BSA (17.7µM) + 8.5mL of water (2hours in the oven at 80°C)
  • Gold/BSA ratio = 166
  1. Solution B: 0.1mL of HAuCl4 (2.9mM) + 0.1mL of BSA (17.7µM) + 0.8mL of water (2hours in the heatblock at 80°C)
  2. Solution D: 1mL of HAuCl4 (2.9mM) + 1mL of BSA (17.7µM) + 8mL of water (2hours in the oven at 80°C)

Data

  • For the heatblock experiments, we obtained:
  1. Some purple fibers into a clear solution for the 166 ratio(B)
  2. A purple solution for the 70 ratio (A)

  • For the oven experiments, we obtained:
  1. A purple solution for the 70 ratio (C)
  2. Some dark purple/black fibers into a clear solution for the 166 ratio (D)
