Objective

Today's objective is to analyze precipitates that formed after performing the first and second dye reactions using dry fluorescence spectroscopy. These analyses will also us to test the presence/absence of dye in the pellet; the presence of dye in the pellet could lead to falsify the dye/BSA ratio calculations.

Protocol

  1. Filled plastic centrifuge tubes with the 166 and 70 Au/BSA ratio dye solutions, and the BSA/HCl dye solutions from both the first and second dye reactions.
  2. Centrifuged all solutions ( 13500 rpm, 5 min).
  3. Poured supernatant from solutions into their respective tubes leaving only the pellets in the plastic centrifuge tubes.
  4. Scraped pellet into separate test tubes.
  5. Pellet was placed onto silica plate, which was prepared for dry fluorescence.
  6. Take fluorescence spectra of sample.
