Objective

Characterize the previously synthesized BSA-L-cysteine ZnS:Mn Nanocrystals using UV-Vis.

Protocol

  1. Remove 1 mL aliquot from test tube and place in the cuvette.
  2. Take the UV-Vis spectra of the sample.
  3. Take the fluorescence spectra of each sample
  4. Repeat for each test tube
  5. Take the UV-Vis spectra of each standard: BSA, L-cysteine, zinc (II) nitrate and manganese (II) acetate.
  6. Data analysis of the UV-Vis absorbances, and fluorescence spectra
  1. Determine if any changes in procedure need to be made.
    1. Is a different scale of protein:ligand ratio required?
    2. Were QD NCs successfully synthesized?
  2. Prepare stock solutions for next synthesis on the following day.

Note: Much of the protocol for today will be determined by the results of our first synthesis.

  • we are going to increase the final volume of solution to yield more mass to be collected. The original final volume will be multiplied by three.

Notes

  • Add before and after photos, and get uv-light photo.

The solutions appeared clear with the formation of white precipitate at the bottom of every test tube. 0% BSA had the most, while 100% BSA had the least. The precipitate emitted orange light when in the presence of ultra-violet rays.
