Objective

Today's objective includes the following:

  1. To prepare KBr Pellets for analysis of our capped nanoparticles using FT-IR.
  2. To prepare samples for analysis using ICP

Protocol

FTIR potassium bromide pellets were made using the following procedure:
250 mg of KBr was dissolved in approximately 25mL of 11-mercaptoundacanamide, 11-mercapto-1-undecanol, lysozyme, and citrate gold nanoparticles. The samples were frozen with liquid nitrogen and lyophilized for two days. The samples were then pelleted and analyzed using Bruker Alpha FT-IR Spectrometer.

ICP was performed using the following:
Standards of gold, sulfur, and the ions in the spring water were made in order to run ICP. 1, 5, 10, 20, and 30 ppm ICP ion standard solution was diluted with 3.0 M nitric acid (HNO3). 1, 7, 14, 21, 28, and 35 ppm gold standard solutions and sulfur standard solutions were diluted with HNO3. Each standard was 30 mL. Standards are run on the ICP first, in order to create a calibration curve. 25% dilution samples at of 15 mL before and after plant absorption are run.
