TAR cloning setup

  • Performed recombinational cloning on uracil dropout plates (blobel lab) with two 50 ml aliquots of chemically competent yeast prepared that morning from a 1:100 dilution of an O.N culture at 30C. 5 hours of incubation yielded cells of OD 0.4 for preparation.
  • 200ul of competent yeast were prepared per 50ml culture and plated onto the uracil dropout plates.
