I re-checked the double restriction on an agarose gel, but it came out the same:



So I re-did the double restriction with PSTI and EcoRI as follows:
Reactive.....1x [ul]
DNA...........5
BSAa..........1
Buffer2......2
H2O..........10
EcoRI........1
PSTI..........1
Total.........20
