I checked the ligations that I made yesterday, this time there are colonies and they seem to be the successful ligations this time, so I replated the 2 ligations od cI plasmid + GFP, for the case of the cI-PCR + GFP + Plasmid 18 I did not re plated them because there wasn’t neither any more petri boxes of LB with Tetracycline nor enough antibiotic to make new boxes, so I just marked the colonies visible. | |||

Saturday 26th June 2010I checked the ligations that I made yesterday, this time there are colonies and they seem to be the successful ligations this time, so I replated the 2 ligations od cI plasmid + GFP, for the case of the cI-PCR + GFP + Plasmid 18 I did not re plated them because there wasn’t neither any more petri boxes of LB with Tetracycline nor enough antibiotic to make new boxes, so I just marked the colonies visible.
