I have been studying the molecular mechanisms underlying oncogenic alterations of cellular metabolism by the c-Myc oncoprotein [http://www.myc-cancer-gene.org (Myc Target Gene Database).] c-Myc heterodimerizes with Max to bind specific DNA sequences (CACGTG), termed E box and transactivates its target genes. Using phylogenetic footprinting and chromatin immunoprecipitation (ChIP) assays, I found that c-Myc overexpression activates genes encoding proteins involved in glycolysis and contributes to the Warburg effect or aerobic glycolysis, which is characteristic of essentially all solid tumors [http://mcb.asm.org/cgi/content/full/24/13/5923?view=long&pmid=15199147 ''(Kim. J. et.al. MCB, 2004'').] I also have been studying metabolic reprogramming of cells in response to hypoxia. I found that hypoxic induction of pyruvate dehydrogenase kinase 1 (PDK1) actively suppresses mitochondrial functions, which results in preserving oxygen consumption and attenuating ROS production under hypoxic conditions [http://www.cellmetabolism.org/content/issue?volume=3&issue=3 (''Kim J. et. al. Cell Metabolism, 2006'').]This metabolic shift confers optimization of efficient respiration of hypoxic cells, which contributes to tumor progression under hypoxic microenvironment. These studies lead to the concept that tumor bioenergetics derived from either cell autonomous oncogenic events or adaptive resopses to hypoxic microenvironment could be an attractive target for the cancer therapeutics.
