It was decided that, given the minimal growth, it would be best to leave the plates in the incubator until Saturday to enable further growth.

I'll pick colonies off of the isolation plates on 4 May 2013 (Saturday) and incubate them for a further week, as this aerobic LB agar environment is hardly the ideal.
 
Thus on 11 May 2013 (next Saturday) I should be able to get here early and do the PCR, digest and gel throughout the day. Primers will need to be ordered.
