Objective

Run DNA gel with test QuikChange from yesterday Prepare media and buffers for re-growth of cells QC with pMXB10 and more cycles

Bench work

  1. Ran 1% agarose DNA gel with pTXB1 QuikChange test from yesterday
    • Lane 1: Ladder, 10μL
    • Lane 4: pTXB1 QC +His 8/18, 5μL sample + 1μL loading buffer
    • Lane 7: pTXB1 QC +His Stage 1, Forward 10cycles 8/18, 5μL sample + 1μL loading buffer
    • Lane 10: pTXB1 QC +His Stage 1, Reverse 10cycles 8/18, 5μL sample + 1μL loading buffer
  2. 2-stage QuikChange protocol with pMXB10
    • Used most recent dilution to 10ng/μL, 8/15/11
    • 12 cycles in stage 1, 18 cycles in stage 2, 8 minute extension step
