==Continuation of pH and Conductivity from 9/16/14==
 
Today we continued the pH and conductivity measurements of our dilute Glycine solutions. The data can be found in my lab entry from [http://openwetware.org/wiki/User:Khyra_A._Neal/Notebook/Chem_571/2014/09/16 September 16, 2014].
 
==Electrophoresis==
 
1. Prepare electrophoresis samples and refrigerate until next week
 
* 50 μL of 1g/L Lysozyme stock solution and dilute to 5 mL with 50 mM Glycine stock solution in volumetric flask.
** Take 10 μL of diluted solution (Lysozyme stock and glycine solution) and 10 μL of running buffer (prepared by Dr. Fox) and add to 1.5 mL centrifuge tube.
**Close lid, vortex for 5-10 seconds, and store in refrigerator.
 
* 50 μL of 30:1 [Au]:[Lysozyme] colloid solution that was prepared in previous Kinetics experiment on [http://openwetware.org/wiki/User:Khyra_A._Neal/Notebook/Chem_571/2014/09/09 September 9, 2014] and dilute to 5 mL with 50 mM Glycine stock solution in volumetric flask.
**Take 10μL of diluted solution and 10μL of running buffer solution and add to 1.5 mL centrifuge tube.
**Close lid,  vortex for 5-10 seconds, and store in refrigerator.
 
* 50 μL of 30:1 [Au]:[BSA] colloid solution prepared on August 26, 2014 and dilute to 5 mL with 50 mM Glycine stock solution in volumetric flask.
**Take 10 μL of diluted solution and 10 μL running buffer and add to centrifuge tube.
** Close lid, vortex for 5-10 seconds, and store in refrigerator.
 
* 50 μL of 1g/L BSA stock solution (prepared by Dr. Fox)  and dilute to 5 mL with 50 mM Glycine stock solution in volumetric flask.
** Take 10 μL of diluted solution (BSA and glycine solution) and 10 μL of running buffer and add to 1.5 mL centrifuge tube.
**Close lid, vortex for 5-10 seconds, and store in refrigerator.
 
*Prepare Soy Protein Solution
** Dissolve 0.00209 g Soy Protein in 2 mL of H<sub>2</sub>O in volumetric flask.
*** 50 μL of soy protein solution was diluted to 5 mL with 50 mM Glycine stock solution
*** Take 10 μL of diluted solution (soy protein and glycine solution) and 10 μL running buffer and add to 1.5 mL centrifuge tube.
***Close lid, vortex for 5-10 seconds, and refrigerate.
 
There are a total of 5 solution that are ready for electrophoresis next week.
 
==Dialysis Continuation==
 
The dialysis samples from yesterday were place on a shaker at low speed and were dialyzed overnight. We removed the samples from the dialysis tubing and stored them in falcon tubes until next week for Bradford assay analysis. The soak solutions were also saved by recovering beakers with parafilm and stored in drawer until next week.

Continuation of pH and Conductivity from 9/16/14

Today we continued the pH and conductivity measurements of our dilute Glycine solutions. The data can be found in my lab entry from September 16, 2014.

Electrophoresis

1. Prepare electrophoresis samples and refrigerate until next week

  • 50 μL of 1g/L Lysozyme stock solution and dilute to 5 mL with 50 mM Glycine stock solution in volumetric flask.
    • Take 10 μL of diluted solution (Lysozyme stock and glycine solution) and 10 μL of running buffer (prepared by Dr. Fox) and add to 1.5 mL centrifuge tube.
    • Close lid, vortex for 5-10 seconds, and store in refrigerator.
