Concentration and Size Determination of AuNPs

The method to determine the size and concentration of the nanoparticles was described in 'Determination of Size and Concentration of Gold Nanoparticles from UV/Vis Spectra' by Wolfgang Haiss

  • Measure the absorbance of the 30:1 Au:Lys colloid to determine the surface plasmon resonance peak (SPR).
  • Run UV/Vis between 800 - 400 nm
  • The SPR peak was found to be 529 nm.
  • The ratio of the absorbance at the SPR peak (A529) and absorbance at 450 nm (A450) was used to determine the diameter of the nanoparticles.
  • The diameter found was used to determine the molar absorptivity at 450 nm (ε450)
  • The concentration was calculated using: c= A450 / ε450
