Objective

  • synthesize AUNP at a concentration of 60 HAUCl4 to 1 BSA
  • prepare stock solutions

Description

  • Stock Solutions
    • HAuCl4 in 50mL solution
      • 2.7mM= 0.0027mols/L x 0.05 L = 1.35E-4 mols HAuCl4
      • 1.35E-4 mols HAuCl4 x 337.79 mols = 0.0458789 grams of HAuCl4
    • BSA in 50mL solutions
      • 0.0000015mols/L x 0.05L = 7.5 E-8 mols
      • 7.5 E-8 mols x 66,000 g/mol = 0.00495 grams of BSA


  • BSA/AuNP Preparation of 60:1 based off stocks above
    • In oven for four hours


Calculations for dilutions of 60:1 in 10 mL

  • (1mLBSA)(0.0000015mols/L)= (10mL)(x)
    • x=0.00000015mols/L of BSA in 10mL of water total volume
  • (60)/(1)=(x)/(0.00000015mols/LBSA)
    • x=0.000009mols/L of HAuCl4
  • (x)(0.0027mols/L)=(10mL)/(0.000009mols/L)
    • x= 0.033mL of HAuCl5 in 10mL of water total volume


  • So,
    • 33µL of HAuCl4
    • 1000µL of BSA
    • 8967µL of H20

Check on ratio of concentration= (0.000009mols/L)/ (0.00000015mols/LBSA) = 60/1
