Objective

Execute procedures from from Dr Harting's AU Biomaterials Design Lab Notebook

Description

BSA-AuNP Procedure

[HAuCl4]=2.54nM<br.> [BSA]=15.6μM<br.> final [HAuCl4] 90x [BSA]→15.6μM×90=1404μM=1.404mM=final [HAuCl4]<br.> (2.54mM HAuCl4 initial)(1mL solution)=(1.404mM HAuCl4 final)(V)<br.> V=1.809mL BSA<br.> Combine 1mL HAuCl4 with 1.810mL BSA.<br.> Rest of procedure followed as written.

Citrate-AuNP Procedure

[HAuCl4]=0.249Mm<BR.> Sodium citrate=1.01%<br.> Rest of procedure followed as written.<br.> Final volume AuNPs: 26.5mL<br.> Absorbance measurements taken of 10x diluted solution AuNPs rather than original sample concentration.<br.>

Data

Absorbance spectra for AuNPs synthesized by citrate reduction.<br.> <br.> <br.> <br.> λspr=518.5nm<br.> Aspr=0.143<br.> A450=0.091<br.> Aspr/A450=0.143/0.091=1.57<br.> According to the Supporting Information from 'Determination of Size and Concentration of Gold Nanoparticles from UV/vis Spectra':<br.> AuNP diameter=12nm<br.> ε450 for 12nm AuNPs=1.09E+08<br.> Using Beer's Law to calculate concentration:<br.> C=1.312E-09M or 1.312nM<br.> Accounting for the 10x dilution factor used in taking absorbance measurements:<br.> C=13.12nM<br.>
