Objective

Run SDS-PAGE to observe the amount of hemoglobin leftover from yesterday.

Description

Gel Electrophoresis: Lane Assignments

  1. 100x Diluted Hemoglobin & Dye
  2. Pepsin 1
  3. Pepsin 2
  4. Pepsin 3
  5. Pepsin 4
  6. Pepstatin 1
  7. Pepstatin 2
  8. Pepstatin 3
  9. Pepstatin 4
  10. 100x Diluted Hemoglobin & Dye
  11. Dr. Fox's sample
  12. 100x Diluted Hemoglobin & Pepsin

JMC's 12. 100x Diluted Hemoglobin & Pepstatin

UV-vis Sample Prep

  • Combine 0.75mL 1M perchloric acid with 0.75mL hemoglobin&pepsin and 0.75mL hemoglobin*pepstatin samples that sat overnight
  • Allow reaction to proceed for one hour (until 2:16pm)
  • Centrifuge for 15 minutes
  • Take supernatant, run absorbance measurements
