Ran a trial in OceanView for Absorbance readings.
 
Run at 70°C set through Qpod
 
#Run A Wizard
#Select spectroscopy
#JAZ is the correct hardware device. Click next
#Select absorbance and click next
#Select integration time as 1 ms
#Select scans to average 10
#Click next
#Open lamp. Click lightbulb to store reference spectrum. Click next
#Close lamp. Click the lightbulb to store dark spectrum. Click finish
#Open lamp. Click save graph to files. Configure
#Save the first available scan every 1 minute
#Stop after 2 hours
#Save to directory in a dropbox folder - 02-20-H2O-70
#File format- Tab separated with headers
#Create file name -  mb_opd_acetate_h202_h20_70_
#Click apply
# When ready to run, click play button, then click the save graph to files button a second time.
 
Cuvette at start time contains-
# 0.675 mL (2.8 mg OPD in 2.8 mL H2O)
# 0.75 mL dilute H2O2 (0.1135 μL H2O2 and 0.8865 μL H2O)
 
First reading taken after 45 seconds.
 
0.225 mL of Mb added after 2 minutes (between readings 2 and 3) (1 mg Mb/KCl/Acetate buffer in 1 mL H2O)
 
Steps were repeated at same temperature with 1 mg Mb/KCl/Citrate buffer in 1 mL H2O
