check competent cells - negative control

  • use competent cells only as negative control, dissolve in 3 ml LB and plate the following dilutions on LB kan/spec plates: 2-5-50μl + spin down, dissolve in 150μl and plate everything → check on mon

use this protocol - link still missing

result from 4/24 - empty pRedroot transformation = positive control

  • transformation of electrocompetent cells with empty pRedroot worked fine no matter which amount was plated → use 5 and 50 µl for the future (agro and ec2 electroporator programs both fine)
