Happy boys and happy girls


  • Today, I arrived early to grow the strains that were grown from the transformation. I cultured them in 4ml of LB with 4μl of ampicilin and I left them growing for ab out 6 hrs. After that, I plated them and extracted plasmid.


  • I left a restriction for a total reaction of 20μl with ECO RI and PST I to check whether strains contain the correct ligations or not. The strains obtained were labeled as 1.1, 1.2, 1.3, 1.4, and 2.1.
