Objective

Acquire fluorescence scans of lysozyme (pH 6) and myoglobin (pH 5) at 80C.

Lysozyme Fluorescence

3 mL Sample

  1. 0.5 μM BSA
    1. Stock Concentration 210 μM
    2. Volume Added: 7.1 μL
  2. pH 6
    1. Stock Concentration 1 mM HCl
    2. Volume Added: 3 μL
  3. Volume water added: 2990 μL

UV-Vis of sample

Evidence here of photobleaching of the fluorophore

Fluorescence scans

  1. Acquisition information
    1. Excitation: 290 nm, 10 nm slit width
    2. Emission: 310-540 nm, 10 nm slit width
    3. 100 nm/minute
    4. 61 measurements, 180 second delay between measurements

Myoglobin Fluorescence

3 mL Sample

  1. 12.5 μM BSA
    1. Stock Concentration 160 μM
    2. Volume Added: 234 μL
  2. pH 5
    1. Stock Concentration 1 mM HCl
    2. Volume Added: 30 μL
  3. Volume water added: 2736 μL

UV-Vis of sample

Evidence of protein aggregation (cloudy solution)

Fluorescence scans

  1. Acquisition information
    1. Excitation: 290 nm, 10 nm slit width
    2. Emission: 310-540 nm, 10 nm slit width
