Objectives

  • Conduct UV-vis analysis for NaMT-CuSO4 exchanged filtrate samples to determine Cu+2 concentration.
  • Determine the concentration of tetra(4-sulfonatophenyl) porphine dihydrogen chloride (porphine) at which UV-vis absorbance is less than 1.
  • Make x films with tetra(4-sulfonatophenyl) porphine dihydrogen chloride (porphine).

UV-Vis of Porphine

  • A 9.445 μM solution of porphine in water was made by adding 1 mL of 94.45 μM porphine (made by Dr. Fox) to 9 mL of water in a tinted Falcon tube.
  • Then, 10 mL each of three ten-fold dilutions of the 9.445 μM porphine solution were made according to the following calculations:
  Solution of 944.5 nM porphine
  (9.445 μM)(V1)=(0.9445 μM)(10 mL)
  V1 = 1 mL of 9.445 μM porphine
  Solution of 94.45 nM porphine
  (0.9445 μM)(V1)=(0.09445 μM)(10 mL)
  V1 = 1 mL of 944.5 nM porphine
  Solution of 9.445 nM porphine
  (94.45 nM)(V1)=(9.445 nM)(10 mL)
  V1 = 1 mL of 94.45 nM porphine
  • Ultimately, 10 mL solutions of 9.445 nM, 94.45 nM, and 944.5 nM porphine were made.


  • Using a Shimadzu UV-2550 spectrophotometer, spectra of all four concentrations of porphine solutions were taken.
    • The range was 200 to 800 nm and the slit width was 1.0 nm.

  • The baseline was conducted with nothing in the cell.
  • For solutions at concentrations 944.5 nM and 94.45 nM, two peaks were visible at 410 nm and 430 nm. It appears that 9.445 nM porphine is not concentrated enough to produce any peaks.
  • The 944.5 nM solution displayed several more peaks past 500 nm, while the 94.45 nM solution did not. Based on the spectra of the solutions, it appears that 944.5 nM porphine will produce the most useful data.
