Objectives

  • With UV/Vis, see how varying concentrations of Tris at pH=10 affect dissolution of HAuCl4 and BSA fibers
  • Prepare solution of HAuCl4 and BSA at a concentration that allows for dissolution rather than precipitation of fibers. The particles in solution will be mixed with Tris buffer to see how they are affected.

Procedure

  1. Measure absorbancy of gold nanoparticles in solutions with varying concentrations of Tris buffer
    1. Solutions with a 170 mole ratio of HAuCl4 and BSA were put in a centrifuge at 3000rpm and 10oC for 5 minutes.
      1. Water was removed with a pipette
    2. Tris stock was prepared
      1. 1mol Tris /1L = 1M
      2. 121.14g/mol Tris
      3. 12.114g Tris was put in 100mL to make 1M stock solution
      4. 9mL water was added to each tube and a serial dilution was done with Tris in 8 tubes.
    3. Absorbance was measured in each of the 8 tubes 3 times using UV/Vis
  2. Solution was made with a mole ratio of 70
    1. 2.4268M BSA solution was used
      1. m1v1=m2v2
      2. 24.266μM * (volume) = 1μM * 6mL
      3. .247mL BSA solution
    2. 2150μM HAuCl4 solution was used
      1. 2150μM * (volume) = 6mL *(1μM*70)
      2. .195mL HAuCl4 solution
    3. 6-.195mL BSA -.247mL HAuCl4 = 5.558mL H2O was added
  3. Solutions went in oven at 85oC from 2:32-6:32

Discussion

  • The three sets of spectra for Tris show almost no change in absorbancy. There is a drop in absorbancy for 1*10-4M Tris solution, but not in any others. More trials should be run to determine if this was due to noise or a change in dissolution.
  • Abigail E. Miller 14:44, 17 September 2012 (EDT):see previous entries for notes on embeding graphs and describing calculations. more details needed in general. these are not trials you ran but time points. how did the solution change as a function of time. at 1 hour after adding tris versus after 24 hours or after a week?
