Enough Lysozyme was added to three test tubes each containing 0.5 mL HAuCl<sub>4</sub> to make [Au]:[Lysozyme]=30 (see chart below for prescription to make such a solution).  The test tubes were covered with foil and placed in an 80°C hot water bath. Every 30 minutes half of the solution in one of the test tubes was poured into a plastic cuvette and placed in the UV-Vis Spectrometer. The UV-Vis from 800 nm to 360 nm was recorded and the above procedure was performed every 30 minutes until no more solution remained.

Colloid v. Fiber Formation

  • Creating a 125 μM stock solution of Lysozyme
    • 50 mL Lysozyme solution, (1.25x10-4 M)(0.050 L)(14300 g/mol)
      • 0.0894g Lysozyme in 50 mL Volumetric Flask
  • Creating a 2.512 mM stock solution of HAuCl4
    • 50 mL HAuCl4 solution, (2.5x10-3 M)(0.050 L)(340 g/mol)
      • 0.043 g HAuCl4 in 50 mL Volumetric Flask

Six solutions of differing [Au]:[Protein] ratios were made with ratios from 10:1 to 60:1. 0.5 mL of HAuCl4 was always used and differing amounts of protein. Water was added to make each solution volume 5 mL. See chart below for volumes of solutions used. The test tubes in which the solutions were combined were then placed in an oven a 80°C for 3 hours.
