The experiment that was performed on August 27th was repeated using 20 minute intervals between measurements.  The same general [[User:Michael_S._Bible/Notebook/571/2014/08/27|procedure]] was followed except that 5 tubes of solution were prepared so that there would be enough to perform 10 measurements over 3 hours.  The results of that experiment can be found below.

Objective

Today we took UV/Vis spectra of the amino acid solutions made on 9/3.

Description

  • 2-3 mL samples of each of the amino acid stock solutions were loaded into polystyrene cuvettes, one at a time, and the data gathered was used to create the graphs below.

Fluorometry

Below is a graph of the data acquired from fluorometry on 81.2 nM L-Tryptophan. A wavelength of 275 nm was used to excite the solution. The small molarity is the result of a 2500:1 dilution of a 203 μM solution of L-Tryptophan. Tryptophan was the only one of our 5 amino acids that produced any significant results in UV-Vis and was, therefore, the only amino acid that was tested using Fluorometry.
