Objective

Today we took UV/Vis spectra of the amino acid solutions made on 9/3.

Description

  • 2-3 mL samples of each of the amino acid stock solutions were loaded into polystyrene cuvettes, one at a time, and the data gathered was used to create the graphs below.

UV-Vis Absorbance Curves for Amino Acids

Fluorometry

Below is a graph of the data acquired from fluorometry on 81.2 nM L-Tryptophan. A wavelength of 275 nm was used to excite the solution. The small molarity is the result of a 2500:1 dilution of a 203 μM solution of L-Tryptophan. Tryptophan was the only one of our 5 amino acids that produced any significant results in UV-Vis and was, therefore, the only amino acid that was tested using Fluorometry.

New Kinetics Experiment

The experiment that was performed on August 27th was repeated using 20 minute intervals between measurements. The same general procedure was followed except that 5 tubes of solution were prepared so that there would be enough to perform 10 measurements over 3 hours. The results of that experiment can be found below.

Results of Kinetics Experiment

Our lysozyme-gold solution, as before, took far longer to show signs of AuNP formation than did the solutions of other groups performing the same experiment. For example, see Paul's results for the same experiment.

See a graph below of the kinetics run for AuNP-formation at 70°C, sampled at 20 minute intervals below:
