Stock Solution Preparation

1. 100 mL of 2.5 mM HAuCl4 solution was made from 0.08539 g of HAuCl4 (MW 339.785 g/mol) in 100 mL of distilled H2O

2. 100 mL of 20 μM lysozyme solution was made from 0.02861 g of lysozyme (MW 14300 Da) in 100 mL of distilled H2O

AuNP Fiber Preparation

50 - 5 mL tubes of 45:1 [Au]:[Lysozme] AuNP solution were prepared with final concentrations of 0.25 mM and 5.56 μM of Au and lysozyme, respectively. Each tubed contained the following:

  • 0.5 mL of 2.5 mM HAuCl4
  • 1.39 mL of 20 μM lysozyme
  • 3.11 mL of distilled H2O

The AuNP solutions were placed in the oven at 80°C for four hours.

Protease and Buffer Preparation

1. 8 samples of approximately 4 mg of pepsin were prepared.

  • 6. 0.00465 g
  • 7. 0.00780 g
  • 8. 0.00589 g

2. 1000 mL of 5 mM Tris/0.2 M NaCl buffer was prepared from 11.68960 g of NaCl and 0.78922 g of Tris in approx. 1000 mL of distilled H2. 0.5 N HCl was added dropwise to a pH of 8.067.
